Below is a guide on how to use an autoclave.

Summary: Autoclave Usage: 1. Adding Water: First close the drain valve, adjust the main valve to "full drainage", then open the water inlet valve and add distilled water to the steam generator. When the water level is about 1-2 cm from the top of the water gauge, close the water inlet valve and set the main valve to "off".

Autoclave Usage:

1. Adding Water: First close the drain valve, set the main valve to "full drain", then open the water inlet valve and add distilled water to the steam generator. When the water level is about 1-2 cm from the top of the water gauge, close the water inlet valve and set the main valve to "off".

2. Loading: Place the items to be sterilized into the chamber. Do not overpack. Flasks and test tubes containing media should be upright or slightly tilted to prevent contamination during sterilization.

3. Closing the Door: Rotate the locking handle clockwise to the red arrow. The handle should engage with the door ring. Then rotate the octagonal turntable to close the door and gasket to prevent leakage during sterilization. Do not overtighten to avoid damaging the gasket.

4. Power On and Heating: When the power control switch is turned to the "ON" position, the power indicator light will illuminate, indicating that the power is on. Then, according to the required sterilization steam pressure of the items, adjust the rotary knob to 0.07 MPa, 0.1 MPa, or 0.14 MPa. The electric heating indicator light will illuminate, indicating that heating has begun. (Autoclave Manufacturer)

5. Insulation and Pressure Maintenance: When the steam in the steam jacket reaches the automatically controlled pressure during heating, the electric heating indicator light will turn off, indicating that heating has stopped. As the pressure decreases due to heat dissipation, the electric heating indicator light will turn on again, indicating continued heating. This shows that the pressure controller is working normally. When the steam in the jacket is heated to the selected control pressure, the steam can be introduced into the sterilization chamber for sterilization.

At this time, the condensation valve in front of the steam-liquid separator should be opened slightly, and then the main valve should be set to "sterilization." The steam in the jacket will enter the sterilization chamber through the main valve for sterilization, while the condensate will be automatically discharged through the steam-liquid separator. At this time, the steam pressure in the steam jacket will drop rapidly, while the steam pressure in the sterilization chamber will gradually increase. The thermometer in the sterilization chamber will also rise as the items are heated by steam. When the instrument temperature rises to the required sterilization temperature (generally 121 degrees), start timing the sterilization and maintain this temperature until sterilization is complete.

6. Unloading: After sterilization, immediately turn off the power. Depending on the nature and requirements of the sterilized items, decide whether the steam in the sterilization chamber should be cooled naturally or discharged slowly or quickly. For items that will not be affected by a sudden pressure drop, such as instruments, utensils, solid koji, soil, peat, etc., the main valve can be directly adjusted to "slow exhaust" or "fast exhaust" for sterilization.

When the pressure in the sterilization chamber drops to "0", slowly rotate the door turntable, release the locking handle, and open the door 5-10 cm. After 20-30 minutes, remove the sterilized items. At this time, the sterilized items are dry. For items such as solution culture media, only the main valve should be set to "slow exhaust" to allow the steam in the sterilization chamber to exhaust slowly to prevent the sudden drop in pressure from causing the culture medium to boil violently. Alternatively, after sterilization, close the main valve and allow the sterilized items to cool naturally. When the sterilization pressure gauge drops to "0", adjust the main valve to "slow exhaust" for a few minutes and remove the sterilized items.

7. Continuous Operation: If there are many items to be sterilized and continuous operation is needed, check the water level first. If the water level is sufficient, continuous use is possible. If water needs to be added, set the main valve to "full drain", open the water inlet valve to add water, and then continue operation.
